Just wanted to drop a note and let the forum know that I was able to successfully launch FF3 as a standalone application while using Comodo Firewall WITHOUT fully disabling Defense+.

Here are the steps I took to achieve this... using Windows Vista FYI

1.) Open Comodo Firewall
2.) Click on 'Defense+'
3.) Choose 'Trusted Files'
4.) Click 'Add', Browse to C:\Program Files (x86)\Filter Forge 3\Bin, add ALL .exe files to the whitelist. Click 'Close'
5.) Choose 'Defense+ Settings'
6.) Choose 'Exclusion Control Settings' Tab, at the bottom of the pane, click 'Exclusions'.
7.) Click 'Add', Browse to C:\Program Files (x86)\Filter Forge 3\Bin, add ALL .exe files to the whitelist. Click 'Close'
8.) Close out of Comodo Firewall settings
9.) Launch FF3
10.) PROFIT.

I hope this helps the next lost soul.
